CCK-8 kit

Cell proliferation is the basis of organism growth, development, reproduction and heredity, and is an important life characteristic of organism. Cells usually proliferate by dividing. Cytotoxic is a simple cell-killing event caused by cells or chemicals, which is not dependent on apoptosis or necrosis.

Cell Counting Kit-8, referred to as CCK-8 Kit, an alternative method of MTT assay, is a rapid and highly sensitive detection kit based on WST (water-soluble tetrazolium salt, chemical name: 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2, 4-disulfobenzene)-2H-tetrazolium monosodium salt) widely used in cell proliferation and cytotoxicity. It can be used for drug screening, cell proliferation assay, cytotoxicity assay, tumor drug sensitivity, and other tests.

Product packaging and storage conditions:
Dry and keep in the dark at 4℃, valid for one year (recommended). -20℃ dry and stored in the dark, valid for two years.

Experimental principle:

CCK-8 cell proliferation detection kit is a rapid sensitivity kit based on WST-8 (chemical name: 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2, 4-disulfonyl benzene)), which is widely used in cell proliferation and cytotoxicity detection. The WST-8 is an upgrade of MTT and works as follows: In the presence of electron-coupled reagents, WST-8 can be reduced by the mitochondrial dehydrogenase to produce the highly water-soluble orange-yellow formazan (Figure 1-2), the color of which is proportional to the proliferation of the number of surviving cells and inversely proportional to cytotoxicity. OD values measured at 450 nm wavelength by enzyme-labeled apparatus can indirectly reflect the number of living cells. CCK-8 method is widely used, such as drug screening, cell proliferation assay, cytotoxicity assay, tumor drug susceptibility assay, and biological factor activity detection.

Note:

1. Phenol red and serum did not interfere with the detection of CCK-8 method;

2. The cytotoxicity of this kit is very low, so after adding WST-8 for color development, the best assay time can be found by repeatedly reading with the microplate reader at different times.
